Her career began on Broadway in the 1985 original production of Biloxi Blues. She later received a Tony Award nomination for the 1989 revival of Our Town.

Throughout her career, she has appeared in numerous productions including Adventures in Babysitting, Biloxi Blues, Big Top Pee-wee, The Freshman, and Awakenings.

Penelope Ann Miller was born to Mark Miller and Beatrice Miller in Los Angeles, California. Both her parents worked in the entertainment industry.

Her mother is a costume designer, publicist, and journalist and her father was a television actor and producer. She has two sisters. Her older sister Marisa Miller is also a film actress, and her younger, Savannah Miller, is a social worker.

After her graduation from high school, she attended Menlo College for two years from 1981 to 1983. After, she moved to New York to study theatre at the HB Studio.
